Size on disk

When I look at the properties of a certain partition on our file-server (using samba), there is allways a large difference between the actual size of the files and the Size on disk... meaning that I lose sometimes up to 20% of disk space!!!
I figure this has something to do with the block-size etc when formatting the partition, but up till now I haven't found the right settings...
